IL-24 antibody products

Interleukin-24 (IL-24, also called Mob-5 or Mda-7) is an important member of the IL-10 family of cytokines discovered by Dr. Peng Liang’s laboratory at Vanderbilt University and it signals through two heterodimeric receptors: IL-20R1/IL-20R2 and IL-22R1/IL-20R2 (References found here). His group used comprehensive Differential Display screening and identified over two dozen genes regulated by ras oncogene. They focused on a gene encoding a novel secreted protein designated IL-24, which is expressed by activated T cells as well as other cell types transformed by ras oncogenes. Through a series of biochemical studies including GenHunter's AP-TAG technology, Dr. Liang's group demonstrated that ras oncogenes could not only induce the expression of IL-24, but also its receptor(s) in the transformed cells.

Both IL-24 and IL-20 share the identical heterodimeric receptors. Upon binding to its receptors, IL-24 induces rapid activation of Stat-1 and Stat-3 transcription factors in the JAK-SAT signaling pathway, which appear to play a role in cell survival and proliferation. Under physiological conditions, the major sources of IL-24 are the activated monocytes and T helper 2 cells, whereas the major IL-24 target tissues, based on the receptor expression pattern, are non-hematopoietic in origin and include skin, lung, and reproductive tissues. IL-24 is part of a complex cytokine network implicated in epidermal functions, tissue repairs/wound healing, autoimmune diseases such as psoriasis, and cancer. Thus, the IL-24 signaling pathway may offer a potential therapeutic target for treatment of cancer and other conditions.

Cos cells transiently transfected with a heterodimeric IL-24 receptor were exposed to the IL-24-alkaline phosphatase fusion protein (IL-24-AP). The ligand bound to the cell surface of transfected cells was visualized by the alkaline phosphatase activity under a light microscope (20X magnification) without phase contrast.
